    Overview

    Collagen is the structural protein of skin, tendon, ligament, cartilage and bone matrix, and hydrolysed collagen supplements are that protein enzymatically cut into short peptides of roughly 2-5 kDa. The old objection — that collagen is digested into amino acids like any protein, so eating it cannot direct anything to skin or joints — turned out to be incomplete. Specific di- and tripeptides such as prolyl-hydroxyproline survive digestion intact, appear in plasma, and act as signalling molecules that stimulate fibroblast and chondrocyte activity rather than simply supplying raw material.

    The best-supported outcome is skin. Meta-analyses of randomised trials at 2.5-10 g/day for eight to twelve weeks show improvements in elasticity, hydration and dermal collagen density, with modest reductions in wrinkle depth. Joint data are next: 5-10 g/day reduces activity-related joint pain in athletes, and 40 mg/day of undenatured type II collagen — a completely different, immune-tolerance mechanism — has separate evidence in osteoarthritis.

    The connective-tissue application is the most interesting recent development. Fifteen grams of collagen with vitamin C taken 30-60 minutes before loading exercise increases collagen synthesis markers, which is the basis for tendon and ligament rehabilitation protocols. Note that collagen is low in leucine and lacks tryptophan, so it should never count toward your protein target for muscle-building purposes.
