- Evidence is insufficient, and the goal is questionable. No human NMN trial has demonstrated a meaningful change in insulin-like growth factor 1. The trials at 250-1,000 mg/day over 6-12 weeks measured blood NAD+, insulin sensitivity, aerobic capacity and walking speed. More importantly, the direction of benefit is unclear: reduced IGF-1 signalling is one of the most reproducible longevity mechanisms across model organisms, with reduced growth hormone and IGF-1 signalling extending lifespan in worms, flies and mice, and people with Laron syndrome, who lack growth hormone receptor signalling, show strikingly low rates of cancer and diabetes. Higher IGF-1 in humans is associated with increased risk of prostate, breast and colorectal cancer. So raising IGF-1 sits awkwardly alongside the longevity framing NMN is usually sold with.
