- Evidence is insufficient. A small number of human trials have measured IGF-1 as a secondary outcome with inconsistent results - one study in healthy older adults reported reduced IGF-1 and IGFBP-3 at high dose, others found no change. The interpretation is also contested: lower IGF-1 is associated with longevity in model organisms and in Laron syndrome, where growth hormone receptor deficiency confers apparent protection from cancer and diabetes, but low IGF-1 in older adults is associated with frailty, sarcopenia, reduced bone density and increased cardiovascular mortality in observational data. The relationship is U-shaped, so lowering IGF-1 is not straightforwardly desirable.
