    Evidence is preliminary and mechanistic. Spermidine induces autophagy principally through inhibition of EP300 acetyltransferase, a pathway that is largely mTOR-independent - this is an important distinction, because spermidine is often marketed as an mTOR inhibitor when the cell biology indicates it works alongside rather than through that pathway. Some crosstalk exists, and effects on mTOR signalling have been reported in cell and animal models, but no human study has measured mTOR pathway activity after spermidine supplementation, and doing so requires tissue biopsy and phosphoprotein analysis.

    What the studies say

    Spermidine extends lifespan in yeast, flies, worms, and mice, and observational human data links higher dietary spermidine to lower mortality. Human trials so far are small pilots (memory in older adults, safety), with no long-term outcome data.
