Spermidine is a naturally occurring polyamine that declines with age and induces autophagy, and it extends lifespan in yeast, worms, flies and mice - the mouse data being more credible than most longevity compounds, with cardioprotective effects and increased median lifespan reported by Eisenberg and colleagues. Observational human data associate higher dietary spermidine intake with lower mortality. Human trials are small and short: the SmartAge trial of 1.2 mg/day over 12 months in older adults with subjective cognitive decline found no significant cognitive benefit, and no trial has measured an aging endpoint.

    What the studies say

    Animal studies show extended lifespan in yeast, flies and mice, and epidemiological data link higher dietary spermidine intake to lower mortality. Human trials so far cover only safety and small cognitive or cardiac endpoints.

    Spermidine has better preclinical credentials than most longevity supplements - the mouse lifespan and cardiac data are real - but the human evidence consists of observational associations with dietary intake and small trials, one of which was negative on its primary endpoint. Dietary intake from wheat germ, aged cheese, mushrooms, soy and legumes is the food-first route and is where the observational associations come from. The interventions with genuine healthspan evidence remain physical activity including resistance training, not smoking, limiting alcohol, sleep, social connection, controlling blood pressure and diabetes, and preventive screening. Safety of spermidine: doses up to 6 mg/day appear well tolerated in short trials, with no established upper limit and no long-term safety data. The main practical caution is that wheat germ extract contains gluten, so it is unsuitable in coeliac disease or wheat allergy unless certified gluten-free. Polyamines are involved in cell proliferation, and there is theoretical concern about supplementation in active cancer, so anyone with a cancer diagnosis should discuss it with their oncology team. Safety in pregnancy and breastfeeding is not established.
