- Evidence is insufficient and the mechanistic link is indirect. Spermidine induces autophagy chiefly by inhibiting the acetyltransferase EP300, which shifts the cellular acetylation balance toward deacetylation - the same net direction as sirtuin activation, which is why the two are often conflated. Spermidine is not a sirtuin activator: it does not bind or stimulate SIRT1, and the autophagy it induces is largely sirtuin-independent, though some crosstalk has been reported in cell models. No human study has measured sirtuin expression or activity after spermidine, and no clinical assay for sirtuin activity exists.
