- Evidence is insufficient in humans by necessity. Spermidine extends lifespan in yeast, nematodes, flies and mice, with the mouse data showing increased median lifespan and cardioprotection - a stronger preclinical record than most longevity compounds. Human evidence is observational: cohort studies including the Bruneck study associate higher dietary spermidine intake with lower all-cause mortality, an association confounded by the whole grains, legumes and vegetables that supply it. No randomised trial can realistically measure human lifespan, and the human trials that exist measure short-term surrogates and have been largely negative on primary endpoints.

Dietary spermidine extended lifespan in yeast, flies, worms and mice, and in aged mice it preserved cardiac diastolic function, reduced hypertrophy and lowered blood pressure. Benefits required intact autophagy and were absent in autophagy-deficient animals. Human cohort data (Bruneck study) linked higher spermidine intake to lower cardiovascular mortality.

Caveats
Long-term human safety data are limited. Doses in supplements are close to dietary levels — food sources are a reasonable alternative.
