Overview
Hericium erinaceus, or lion's mane, is an edible mushroom whose fruiting body and mycelium contain hericenones and erinacines — compounds that stimulate nerve growth factor synthesis in cell culture and, unusually for a large molecule question, appear able to influence NGF-dependent pathways in vivo in rodents.
The best-known human study randomised 30 Japanese adults aged 50-80 with mild cognitive impairment to 3 g daily of powdered fruiting body or placebo for 16 weeks. The treatment group improved on a cognitive function scale at weeks 8, 12 and 16, and those gains disappeared four weeks after stopping — a pattern that argues for a genuine but non-durable effect. Later small trials have reported reductions in anxiety and depression scores and improvements in sleep quality, though a well-conducted trial in healthy young adults found no acute cognitive benefit.
Lion's mane is also studied for peripheral nerve regeneration, so far only in animals. Its practical appeal is that it is a food with a good safety record: it can be eaten as a mushroom, and supplemental doses are modest. Product quality is the biggest variable — mycelium-on-grain products contain substantial starch filler and far less of the active fraction than fruiting-body extracts.

Very well tolerated. Mild digestive discomfort is the most common complaint; skin rash and respiratory reactions have been reported in people with mushroom allergy.
