    Evidence is insufficient. The rodent literature is suggestive - lion's mane compounds stimulate NGF, and animal studies report improved performance on novel object recognition and maze tasks, consistent with effects on hippocampal memory encoding. The human evidence does not reach this endpoint. The one positive trial, in 30 older Japanese adults with mild cognitive impairment taking 3 g/day for 16 weeks, used a composite cognitive scale rather than a memory encoding measure, and has not been replicated. No adequately powered trial has tested whether lion's mane improves memory formation in healthy adults, and the studies that exist are small, brief and use inconsistent preparations of highly variable potency.

    Rodent studies show improved memory task performance, but human trials are limited to small samples in mild cognitive impairment or with acute dosing. Marketing claims substantially exceed the clinical record.

    Caveats

    For memory encoding, sleep is the intervention with overwhelming evidence - consolidation happens during sleep, and no supplement compensates for restricting it. Spaced retrieval practice and aerobic exercise also have real support. New or progressive memory problems need medical assessment, since B12 deficiency, thyroid disease, depression, sleep apnoea and medication effects are treatable causes. Safety: lion's mane is generally well tolerated at 1-3 g/day over months, with mild gastrointestinal upset most common; skin rash and rare respiratory allergic reactions have been reported, and it should be avoided with mushroom allergy. Possible mild antiplatelet and glucose-lowering activity means caution with anticoagulants, antiplatelets and diabetes medication, and stopping two weeks before surgery. No established upper intake level and no long-term safety data beyond about a year. No safety data in pregnancy or breastfeeding - avoid in both, and in children. Product potency varies widely; mycelium-on-grain products are largely starch.
