    Biotin (Vitamin B7) for Nail Strength

    Overview

    Biotin is the most commonly recommended supplement for brittle nails, and the recommendation is older and firmer than the evidence behind it. A systematic review of biotin for nail disease found the case rests on three small uncontrolled studies with no randomised controlled trials.
    Those studies did report thicker, less splitting nails in a majority of participants taking 2.5 mg daily for several months. Without a placebo arm, that finding cannot be separated from natural fluctuation in nail quality and the slow regrowth cycle itself. The reasonable position is that biotin is cheap, safe and plausibly helpful in brittle nail syndrome, while the evidence remains weak. Anyone with a genuine biotin deficiency, which is rare, is a different and much stronger case.

    Three small uncontrolled studies suggest improvement in brittle nail syndrome at 2.5 mg daily. No randomised controlled trial exists, so the effect size is unverified.

    How It Works

    Biotin is a cofactor for carboxylase enzymes involved in fatty acid synthesis and amino acid metabolism. Nail plate is keratin embedded in an intercellular lipid matrix, and the proposed mechanism is improved lipid synthesis giving a better-cemented, less splitting plate.
    This mechanism is borrowed from veterinary work, where biotin reliably improves hoof horn quality in horses and pigs. The extrapolation to human nails is plausible but has never been demonstrated at the tissue level in a controlled human study. A practical consequence is timing. Fingernails take roughly six months to grow out completely, so any real change appears at the free edge slowly and cannot be judged at a few weeks.

      Fungal infection, psoriasis, thyroid disease and iron deficiency all cause nail changes that biotin will not fix.

    The linked systematic review is the honest summary of this field: three small uncontrolled studies, positive in direction, with no randomised controlled evidence. It is enough to justify a cheap trial in brittle nail syndrome and not enough to promise a result.

    Safety

    Biotin itself is remarkably safe. It is water-soluble, has no established toxic dose, and no upper limit has been set. The real hazard is diagnostic rather than physiological.
