TL;DR
Exercise is by far the strongest known BDNF stimulus. Lion's mane, omega-3 and curcumin have supportive but preliminary human data; serum BDNF is a poor proxy for brain levels.
Why It Matters
Brain-derived neurotrophic factor supports neuron survival, synaptic plasticity and hippocampal neurogenesis, and low levels are associated with depression, Alzheimer's disease and poor cognitive ageing. The important caveat is measurement: serum and plasma BDNF correlate only loosely with brain concentrations and are heavily affected by platelet content, time of day and processing method. Treat published BDNF changes as suggestive, and judge interventions on cognitive and mood outcomes instead.
How to Measure
There is no validated clinical BDNF test. Serum BDNF is used in research but is not actionable individually. Judge progress by outcomes that matter: validated cognitive batteries, PHQ-9 for mood, sleep quality, and cardiorespiratory fitness, which tracks BDNF response more reliably than any supplement.

Optimization Protocol
Aerobic exercise is the primary lever: moderate to vigorous cardio acutely raises circulating BDNF and produces sustained increases with training, with high-intensity intervals showing the largest effect. Protect deep sleep, during which BDNF-dependent consolidation occurs. Add intermittent fasting or caloric restriction, both of which raise BDNF in animal and some human work. Supplement layer, all secondary: lion's mane 500-1000 mg/day, omega-3 1-2 g/day EPA+DHA, bioavailable curcumin 500-1000 mg/day, and correct low vitamin D. Chronic alcohol and chronic stress both suppress BDNF and are worth removing first.
