- Take with food, 1-2 hours before bed. The mechanism has a real biochemical basis: magnesium is a cofactor for enzymes in the pathway converting serotonin to melatonin, including N-acetyltransferase, and magnesium deficiency in animal studies reduces pineal melatonin output. In humans, the 8-week randomised trial in elderly people with insomnia that used 500 mg/day magnesium oxide reported increased serum melatonin alongside improved sleep time, efficiency and onset latency, and reduced serum cortisol - which is the most direct human evidence available. It is one trial in a specific population, melatonin was a secondary outcome, and the effect is unlikely to match taking melatonin directly. Treat this as supporting endogenous synthesis rather than replacing exogenous melatonin.
