- Evidence is insufficient. No randomised trial has examined the effect of magnesium supplementation on DHEA or DHEA-sulphate concentrations. The claim appears to derive from magnesium involvement in HPA axis regulation and from small studies suggesting magnesium influences cortisol and testosterone under certain conditions - one trial in athletes reported increased free and total testosterone with magnesium during exercise training, which is a different hormone with a different regulatory pathway. DHEA is produced by the adrenal cortex under ACTH control, and while steroidogenesis broadly requires magnesium-dependent enzymes, there is no human evidence that supplementation raises DHEA in people with normal magnesium status. Age-related DHEA decline is not a magnesium deficiency.
