    5-HTP for Melatonin Production

5-HTP is the direct serotonin precursor; since melatonin is synthesized from serotonin, it supplies the substrate for pineal production, with sleep-onset evidence in trials.

    Overview

5-HTP is the direct serotonin precursor; since melatonin is synthesized from serotonin, it supplies the substrate for pineal production, with sleep-onset evidence in trials.

    How It Works

5-HTP crosses the blood-brain barrier and converts to serotonin, which the pineal gland acetylates and methylates into melatonin.

    Dosing & Protocol

    Typical dose

    Recommended dose
100–200 mg, 30–60 min before bed; start at 100 mg.
    Expected timeframe
    3-14 nights

    Protocol

    form
Griffonia simplicifolia seed extract, enteric-coated if nausea occurs
    timing
Evening only, in dim light - bright light after dosing suppresses the melatonin step you are trying to feed
    duration
    2-4 weeks
    titration
Stay at 100 mg for one week; only move to 200 mg if there is no nausea and no benefit
    measurement
Track sleep-onset latency in a diary; salivary melatonin testing is optional and rarely necessary
    starting dose
100 mg 30-60 minutes before bed

    Evidence

    What the studies say

RCTs show 5-HTP reduces sleep-onset latency and increases REM sleep. The serotonin-to-melatonin substrate logic is biochemically sound; direct pineal-output studies are fewer.

No studies are yet linked to both 5-HTP and Melatonin Production.

    Safety

    Caveats

Never combine with SSRIs/SNRIs or MAOIs (serotonin syndrome risk); nausea common at higher doses.

    Less likely to help if

People on serotonergic medication cannot use it; primary circadian misalignment needs light therapy, not precursors.
