    How It Works

    Leucine binds Sestrin2 and relieves inhibition of mTORC1, switching on the translational machinery for muscle protein synthesis. That signal is real and rapid. The problem is downstream: building muscle protein requires all twenty amino acids as raw material, and providing only three means the remaining essential amino acids must come from breakdown of existing tissue, which caps net synthesis at roughly half the response to a complete protein dose. BCAAs also compete with tryptophan for blood-brain barrier transport, the proposed basis for the modest reductions in perceived exertion reported in some endurance work.

    What the studies say

    Isotopic tracer studies provide the decisive evidence: infusing or ingesting BCAAs alone produces a muscle protein synthesis response substantially smaller than an equivalent dose of whey, because the other essential amino acids become rate-limiting. A widely cited 2017 review concluded that BCAA supplementation alone cannot support increased muscle protein synthesis in humans. Trials on hypertrophy and strength outcomes are mixed and generally null once total protein intake is equated, and the positive studies tend to involve participants with low baseline protein intake or energy restriction. There is more supportive evidence for modest reductions in delayed-onset muscle soreness and perceived exertion, particularly in untrained participants performing unaccustomed eccentric exercise, though effect sizes are small. Separately, elevated circulating BCAA concentrations are a robust biomarker of insulin resistance in metabolic epidemiology, and while causality is unresolved it is a reason not to megadose chronically.

    Caveats

    The core caveat is opportunity cost rather than harm: the money buys three amino acids when whey buys all twenty plus roughly the same leucine. Chronic high intakes sit awkwardly alongside the BCAA-insulin resistance literature.

    Less likely to help if

    Anyone consuming 1.6-2.2 g/kg of protein daily — which is most people training seriously — will see nothing from adding BCAAs.
