    How It Works

    Withanolides, the active glycosides in Withania somnifera root, appear to act on the hypothalamic-pituitary-adrenal axis at multiple points. Animal and mechanistic work shows dampened corticotropin-releasing hormone signalling and reduced adrenal cortisol output in response to stressors, without the suppression seen with exogenous steroids. A parallel route runs through GABAergic signalling: withanolides have GABA-mimetic activity at GABA-A receptors, which plausibly explains the anxiolytic and sleep effects reported alongside cortisol change. Antioxidant and anti-inflammatory actions in the adrenal cortex may protect steroidogenic tissue from chronic stress load, though this is less firmly established.

    Safety

    Rare cases of ashwagandha-associated liver injury have been reported, usually reversible on stopping. Stop and seek advice for jaundice, dark urine or unexplained fatigue. Ashwagandha is also traditionally abortifacient and should be avoided in pregnancy.
