    Sulforaphane for Phase II Detox Support

    Sulforaphane is the best-documented dietary activator of phase II detoxification enzymes, and randomized trials show it measurably speeds excretion of airborne pollutants in humans.

    Overview

    Phase II detoxification is the conjugation step: the liver attaches glutathione, sulfate or glucuronide groups to reactive intermediates so they can be excreted. Sulforaphane is the best-evidenced dietary compound for upregulating this machinery in humans.
    The linked trial is unusually direct for a supplement claim. Rather than measuring an enzyme in a dish, it measured urinary excretion of conjugated benzene and acrolein in people breathing polluted air, which is phase II activity showing up as an actual output.

    How It Works

    Sulforaphane works by disabling a brake. Under normal conditions the protein KEAP1 keeps the transcription factor NRF2 marked for destruction. Sulforaphane modifies cysteine residues on KEAP1, NRF2 escapes and moves to the nucleus, and it switches on the antioxidant response element.
    Downstream of that element sit the phase II genes: glutathione S-transferases, NAD(P)H quinone dehydrogenase 1, glutamate-cysteine ligase and UDP-glucuronosyltransferases. Because the effect works through gene transcription rather than direct chemical scavenging, it outlasts the compound's short plasma half-life.

    Dosing & Protocol

    Dosing here is about the precursor, not the compound. Glucoraphanin needs the enzyme myrosinase to become sulforaphane, and myrosinase is destroyed by cooking. Supplements supplying glucoraphanin without active myrosinase yield a fraction of the sulforaphane of a myrosinase-containing product.

    Safety

    Sulforaphane at dietary and supplemental doses has a good tolerability record, and the linked trial reported no significant safety problems. Gastrointestinal complaints, especially with sprout beverages, are the usual issue.
