    Sulforaphane for Oxidative Damage Repair

    Sulforaphane is the most compelling option because it upregulates your own antioxidant enzymes rather than acting as a one-shot scavenger.

    Overview

    Sulforaphane does not repair oxidative damage directly - it switches on the cellular machinery that does, which is a meaningfully different and arguably more interesting claim.
    It is the most potent known natural activator of Nrf2, the transcription factor controlling several hundred cytoprotective genes including those for glutathione synthesis, glutathione S-transferases, NAD(P)H quinone dehydrogenase 1 and heme oxygenase-1. Human trials of broccoli sprout preparations show measurable increases in detoxification enzyme activity and accelerated urinary excretion of airborne pollutant conjugates, most notably in a large Chinese trial in a heavily polluted region. Direct repair endpoints - DNA adduct clearance, 8-OHdG reduction, protein carbonyl normalisation - are far less studied in humans. The upstream activation is well demonstrated; the downstream repair benefit is largely inferred.

    How It Works

    Under normal conditions Nrf2 is held in the cytoplasm by Keap1 and continuously degraded. Sulforaphane modifies reactive cysteine residues on Keap1, releasing Nrf2 to translocate to the nucleus and bind antioxidant response elements in the promoters of protective genes.
    The genes switched on are phase II enzymes rather than direct antioxidants: glutamate-cysteine ligase increases glutathione synthesis, glutathione S-transferases conjugate electrophiles for excretion, and thioredoxin and peroxiredoxin systems are upregulated. Because this is a transcriptional response, the effect outlasts the compound itself - a single dose produces enzyme induction lasting days. Repair proper is handled by base excision repair, nucleotide excision repair and the proteasome. Nrf2 activation supports these indirectly by reducing ongoing damage and improving redox conditions, rather than by driving repair enzymes directly.

    Glucoraphanin without myrosinase is largely wasted

    The precursor needs the enzyme to convert. Choose products supplying active myrosinase, or add a little mustard seed powder, which is a rich myrosinase source.

    The Nrf2 mechanism is exceptionally well characterised, and randomised human trials of broccoli sprout preparations have shown increased urinary excretion of benzene and acrolein conjugates in polluted environments, changes in detoxification enzyme expression, and reductions in some inflammatory markers. This is unusually solid mechanistic evidence for a food-derived compound. Clinical repair outcomes are another matter. Trials are short, small, use varied and often poorly characterised preparations, and rely on biomarkers rather than disease endpoints. No trial has shown that sulforaphane repairs accumulated oxidative damage or alters any hard clinical outcome dependent on that repair.

    Safety

    Broccoli sprout preparations are well tolerated at studied doses. Bloating, flatulence and mild digestive discomfort are the usual complaints, and the sulphurous taste puts some people off more than any side effect does.

    Raw sprouts carry a contamination risk

    The warm, humid conditions used to grow sprouts favour Salmonella and E. coli. Pregnant, elderly and immunocompromised people should avoid raw sprouts.

    Very high intakes of cruciferous glucosinolates can affect thyroid iodine uptake, which matters mainly for people with existing thyroid disease or low iodine intake. Safety data in pregnancy and long-term use beyond a few months are limited, and concentrated extracts have not been studied over years.
