Meta-analyses of randomised trials show EPA-predominant omega-3 produces a small to moderate reduction in depressive symptoms, with the clearest benefit as an adjunct to antidepressants in people with diagnosed major depression, and larger effects in those with elevated inflammatory markers. In people without a depressive disorder, effects on general mood and wellbeing are much smaller and often absent - the large VITAL-DEP trial found no benefit for preventing depression in healthy older adults. Proposed mechanisms include membrane fluidity effects on serotonergic and dopaminergic signalling and reduced neuroinflammation.
