    Evidence is insufficient. No human trial has examined the effect of magnesium supplementation on pregnenolone. Pregnenolone is the first steroid formed from cholesterol by the mitochondrial enzyme CYP11A1, and it sits upstream of progesterone, cortisol, DHEA and the sex hormones. Steroidogenesis broadly depends on magnesium-containing cofactors and ATP, which is where the claim originates, but that is a general statement about cellular metabolism rather than evidence of a specific effect. There is no human data showing that supplementing magnesium in a replete person raises pregnenolone, and pregnenolone levels are not routinely measured or clinically meaningful outside specialist endocrine investigation.
