Abstract
A randomized, double-blind, placebo-controlled study showing that 250mg NMN daily for 12 weeks significantly increased blood NAD+ levels and improved muscle insulin sensitivity in healthy older men, demonstrating safety and efficacy.
Methodology
Randomised, double-blind, placebo-controlled trial with blood NAD+ metabolomics, hyperinsulinaemic clamp-related measures and physical performance testing.
Key Findings
In 42 healthy older Japanese men, NMN 250 mg/day for 12 weeks raised whole-blood NAD+ metabolites and improved gait speed and right-hand grip strength versus placebo, while muscle insulin sensitivity and most other metabolic measures did not differ significantly.
Conclusions
Chronic NMN reliably raises circulating NAD+ and produced small functional gains in muscle performance in older men; broader metabolic benefits were not demonstrated.
Limitations
