TL;DR
AMPK is the cell's low-energy sensor, switching on glucose uptake and fat oxidation. Exercise and energy restriction activate it powerfully; supplement evidence rests mostly on cell and animal work.
Why It Matters
AMPK responds to a rising AMP:ATP ratio by promoting catabolic pathways and suppressing lipogenesis and mTOR signalling. It is the pathway behind much of metformin's and exercise's metabolic benefit. The key caveat for consumers: activating AMPK in a petri dish at high concentrations does not establish that an oral supplement reaches meaningful tissue levels in humans. Human outcome data exist for berberine (glycaemia) but are thin for most other "AMPK activators".
How to Measure
AMPK activity itself is not clinically measurable outside a muscle biopsy. Track downstream metabolic markers instead: fasting glucose and insulin, HOMA-IR, HbA1c, triglyceride-to-HDL ratio, waist circumference, and VO2 or work capacity as a proxy for training adaptation.

Avoid stacking multiple unproven "activators" — cost rises faster than benefit.
