The hypothesis comes from Patrick and Ames, who identified a vitamin D response element in the oxytocin gene promoter and proposed that calcitriol could regulate oxytocin transcription, with relevance suggested for autism and social behaviour. This is a molecular biology observation supported by cell work and some animal data. No human trial has measured plasma or central oxytocin after vitamin D supplementation, and peripheral blood oxytocin is in any case a poor and disputed proxy for brain oxytocin activity, since the peptide crosses the blood-brain barrier poorly in either direction.
