    Saffron for Depression

    Saffron extract at 30 mg/day reduced Hamilton Depression Rating Scale scores as much as fluoxetine 20-40 mg and imipramine 100 mg in head-to-head randomised trials, with fewer side effects.

    Overview

    Saffron has an unusually consistent set of trials for a spice. Across randomised studies in mild to moderate depression, standardised saffron extract outperformed placebo and, in several head-to-head comparisons, performed similarly to low-dose fluoxetine or imipramine. The limitations are real: most trials are small, short and conducted by a handful of Iranian research groups. That is why the verdict is likely rather than strong, and why saffron belongs in a conversation with a clinician rather than as a unilateral swap for prescribed treatment.

    What the evidence supports

    Clinically meaningful reduction in depression scores in mild to moderate depression over 6-8 weeks.

    How It Works

    Crocin and safranal, saffron key constituents, appear to inhibit reuptake of serotonin, dopamine and noradrenaline, while also reducing oxidative stress and inflammatory signalling in brain tissue. Both pathways are plausible contributors to the antidepressant signal.
    The monoaminergic component is what makes combination with prescribed antidepressants a question for a clinician rather than an experiment to run at home.

    Dosing & Protocol

    Nearly every positive trial used 30 mg per day of a standardised stigma extract, usually split into two doses. Higher doses have not shown extra benefit and move towards the toxicity threshold.

    More is not better here. Doses of 5 g and above are considered toxic, and 30 mg per day is what the evidence supports.

    Both analyses point the same way with similar effect sizes. The shared weakness is geographic and methodological concentration: small trials from overlapping research groups, with few independent replications elsewhere.

    Safety

    At 30 mg per day saffron is well tolerated, with occasional nausea, appetite change, headache or dry mouth. Very high doses are genuinely toxic, and saffron has traditional use as an abortifacient, so it should be avoided in pregnancy.

    Depression needs oversight

    Do not stop prescribed antidepressants to try saffron. Any worsening mood or thoughts of self-harm needs urgent professional help.
