    L-histidine is essential in humans and sits upstream of histamine, carnosine and urocanic acid — the last of which is a component of the skin's natural moisturizing factor and its UV-absorbing capacity. That skin link produced the most interesting supplement research: a small proof-of-concept trial in adults with atopic dermatitis found oral histidine improved disease severity scores over four weeks, attributed to restored filaggrin breakdown products in the stratum corneum. It is a genuinely plausible mechanism, but the study was small, industry-adjacent and has not been convincingly replicated, so this remains preliminary. Histidine has also been studied in dialysis patients, where low plasma levels track with inflammation and poor outcomes, and in metabolic syndrome, where a small trial in obese women with metabolic syndrome reported reduced insulin resistance and inflammatory markers. None of this justifies routine supplementation in people eating adequate protein — histidine deficiency is not a practical concern on a normal diet. The main caution is that histidine is the direct precursor to histamine, so people with mast cell activation or histamine intolerance sometimes report symptom flares.
