    GABA is a large, polar molecule with very limited blood-brain barrier permeability. Proposed alternative routes include enteric nervous system signalling and vagal afferents, which remain speculative in humans.

Evidence is mixed and the central problem is mechanistic: GABA is a large polar molecule that crosses the blood-brain barrier poorly, so oral GABA is very unlikely to raise brain GABA levels meaningfully. Small trials have nonetheless reported reduced subjective stress and altered EEG alpha/beta ratios within an hour of 100-200 mg, and the leading explanation is action on the enteric nervous system and vagal signalling rather than direct central effect. Trials are small, short, often manufacturer-funded, and inconsistently replicated.

    What the studies say
