    Is magnesium citrate treatment effective on pain, clinical parameters and functional status in patients with fibromyalgia?

    Methodology

    Controlled trial in premenopausal women meeting ACR fibromyalgia criteria plus 20 healthy controls; three 8-week treatment arms of magnesium citrate 300 mg/day, amitriptyline 10 mg/day, or both; pain intensity and threshold, tender points, FIQ, Beck depression and anxiety scores and serum and erythrocyte magnesium.

    Key Findings

    60 premenopausal women with fibromyalgia and 20 matched healthy controls were assessed, with patients allocated to magnesium citrate 300 mg/day, amitriptyline 10 mg/day, or both, for 8 weeks (20 per group). Serum and erythrocyte magnesium were significantly lower in fibromyalgia patients than controls and correlated negatively with symptom severity. Magnesium citrate alone significantly reduced the number of tender points, tender point index, Fibromyalgia Impact Questionnaire and Beck depression scores, while the magnesium plus amitriptyline combination improved every measured parameter except numbness.

    Conclusions

    The magnesium citrate treatment was only effective tender points and the intensity of fibromyalgia. However, it was effective on all parameters when used in combination with amitriptyline.

    Limitations

    Sixty premenopausal women in three arms of 20 with no placebo group and only 8 weeks of treatment; magnesium alone did not improve all outcomes and the design cannot separate expectation effects.
