    Effect of a herbal extract containing curcumin and piperine on midazolam, flurbiprofen and paracetamol (acetaminophen) pharmacokinetics in healthy volunteers

    Methodology

    Randomized, placebo-controlled six-way crossover study; curcuminoids 4 g plus piperine 24 mg dosed four times over 2 days before probe drugs midazolam (CYP3A), flurbiprofen (CYP2C9) and paracetamol (UGT/SULT)

    Key Findings

    Despite in vitro inhibition signals, the curcuminoid/piperine preparation produced no clinically important change in midazolam or flurbiprofen pharmacokinetics in healthy volunteers.

    Conclusions

    Short-term high-dose curcumin with piperine does not meaningfully inhibit phase I CYP3A or CYP2C9 activity in humans.

    Limitations

    Very small sample (n=8), short dosing period, healthy young volunteers only.
