Methodology
12-week randomized, double-blind, placebo-controlled trial of live A. muciniphila (AKK-WST01) vs placebo alongside routine lifestyle guidance in adults with overweight/obese type 2 diabetes (NCT04797442), with germ-free mouse verification.
Key Findings
Both groups lost weight and reduced HbA1c with no significant between-group differences. Only participants with low baseline A. muciniphila showed high colonization and significant reductions in body weight, fat mass and HbA1c.
Conclusions
Metabolic benefits of A. muciniphila supplementation appear to depend on baseline intestinal levels, supporting microbiota-guided probiotic use.
Limitations
Small sample; subgroup findings are exploratory and were not the pre-specified primary endpoint; the overall trial was null.
Supplements Studied
In participants with low baseline A. muciniphila, AKK-WST01 supplementation showed high colonization efficiency and significant reductions in body weight, fat mass, and HbA1c, which were not found in the placebo group.
