Methodology
Interim 3-year MRI voxel morphometry analysis of 56 participants in the randomized, double-blind, placebo-controlled ASCOMALVA trial comparing donepezil plus placebo with donepezil plus choline alphoscerate in Alzheimer's disease with cerebrovascular injury.
Key Findings
Gray matter atrophy was less pronounced in the donepezil plus choline alphoscerate group in frontal and temporal lobes, hippocampus and amygdala compared with donepezil plus placebo, paralleling less decline on neuropsychological tests.
Conclusions
Adding choline alphoscerate to donepezil appears to partly counter brain volume loss in Alzheimer's disease, with likely functional relevance.
Limitations
Interim analysis of a small single-centre trial, imaging endpoints are surrogate measures, and the population had comorbid cerebrovascular injury.
Supplements Studied
In the D + CA group (n = 29) the gray matter atrophy was less pronounced compared to the D + P group in frontal and temporal lobes, hippocampus, and amygdala.
