    Effects of extended-release niacin with laropiprant in high-risk patients (HPS2-THRIVE)

    Methodology

    Randomised placebo-controlled trial of extended-release niacin with laropiprant in 25,673 adults with vascular disease on statin therapy, followed a median 3.9 years.

    Key Findings

    No reduction in major vascular events, with significant excesses of diabetes-related complications, infection, bleeding and gastrointestinal and musculoskeletal adverse effects.

    Conclusions

    Niacin adds no cardiovascular benefit on statin therapy and causes significant harm.

    Limitations

    Laropiprant co-administration complicates attribution of some adverse events.
