    Abstract

Randomized controlled trial showing myo-inositol 4g daily significantly improved mood symptoms, depression, and anxiety in premenstrual dysphoric disorder through modulation of neurotransmitter systems.

    Methodology

Randomised, double-blind, placebo-controlled trial comparing myo-inositol powder with placebo in women with premenstrual dysphoric disorder over consecutive menstrual cycles, using standardised symptom rating scales.

    Key Findings

Myo-inositol produced greater improvement than placebo in mood, depressive and anxiety symptom ratings in women with PMDD, with good tolerability and no notable adverse effects.

    Conclusions

Myo-inositol may offer modest symptomatic benefit in premenstrual dysphoric disorder, plausibly via second-messenger effects on serotonergic signalling, but the evidence base is small.

    Limitations

Small sample, single centre, short follow-up over a few cycles, subjective self-rated outcomes, and no active comparator such as an SSRI.
