B. longum has produced two of the more credible human gut-brain results in the field: reduced depression scores with measurable fMRI changes (NCC3001) and modest stress reduction in healthy adults (strain 1714). Both rest on small, single-site trials that have not been independently replicated.

B. longum NCC3001 reduced depression scores in IBS patients versus placebo at 6 weeks and altered limbic brain responses, but did not significantly reduce anxiety or core IBS symptoms.

Caveats
Every supporting trial is small, single-site, and commercially connected, and the neuroimaging result has never been independently replicated. Effects require continued dosing because the organism does not durably colonise an adult gut.
