    B. longum has produced two of the more credible human gut-brain results in the field: reduced depression scores with measurable fMRI changes (NCC3001) and modest stress reduction in healthy adults (strain 1714). Both rest on small, single-site trials that have not been independently replicated.

    How It Works

    B. longum ferments oligosaccharides to short-chain fatty acids, principally acetate, which support epithelial barrier integrity and reduce systemic exposure to bacterial products. Proposed CNS routes include vagal afferent signalling, modulation of gut-derived cytokines, and effects on tryptophan metabolism. The human fMRI data showing reduced amygdala and fronto-limbic reactivity give this pathway more substance than the usual rodent-only story.

    Evidence

    What the studies say

    Strain NCC3001 was tested in 44 adults with IBS and mild-to-moderate mood symptoms. Over six weeks, 64% reached a two-point or greater reduction in depression score versus 32% on placebo, and functional MRI showed reduced limbic reactivity to negative emotional stimuli — one of the few human neuroimaging results in the probiotic literature. Strain 1714 has been tested in healthy volunteers under acute laboratory stress. A crossover trial in 22 men found reduced cortisol output and lower subjective anxiety during the cold pressor test after four weeks, with accompanying EEG changes. A later trial in students during exam stress found improvements in perceived stress but not on all measures. Set against this: sample sizes of 22 to 44, single research centres with commercial involvement, no independent replication of the neuroimaging findings, and inconsistent results across the secondary endpoints. Meta-analyses of probiotics for mood find small pooled effects with substantial heterogeneity and probable publication bias. The reasonable reading is that the gut-brain axis is real and B. longum is among the better-supported candidates, but the effect size in ordinary use is likely smaller than the headline trials imply.

    Safety

    Caveats

    Every supporting trial is small, single-site, and commercially connected, and the neuroimaging result has never been independently replicated. Effects require continued dosing because the organism does not durably colonise an adult gut.

    Less likely to help if

    People without gut symptoms and without elevated baseline stress showed the least change. This is not a treatment for clinical anxiety or depression, and should never displace evidence-based care.
