Published in Circulation, Vol. 145 (18), pp. 1387-1397
Methodology
Randomised, double-blind, multicentre trial (AVADEC) in men from the community with an aortic valve calcification score above 300 AU. Daily 720 mcg MK-7 plus 25 mcg vitamin D, or placebo, for 24 months. Primary outcome: change in aortic valve calcification; secondary outcomes included aortic and coronary artery calcification.
Key Findings
365 men, mean age 71. Valve calcification rose by 275 AU on MK-7 plus vitamin D and 292 AU on placebo (difference 17 AU, P = 0.64). Aortic and coronary calcification progression did not differ. Cardiovascular events: 10 vs 10. Inactive matrix Gla protein (dp-ucMGP) fell with treatment (P < 0.001).
Conclusions
Two years of MK-7 plus vitamin D did not slow aortic valve calcification in older men.
Limitations
Men only; MK-7 given with vitamin D, so the effect of K2 alone cannot be separated; imaging outcomes.
Supplements Studied
2 years MK-7 plus vitamin D supplementation did not influence AVC progression
