Methodology
Pooled individual-data analysis of 31 databases (11,239 venous thromboembolism cases, 21,521 controls) on factor V Leiden, prothrombin 20210A and MTHFR C677T.
Key Findings
Homozygous C677T showed no significant association with first venous thromboembolism (OR 1.38, 95% CI 0.98-1.93), while factor V Leiden (OR 4.22) and prothrombin 20210A (OR 2.79) heterozygosity raised risk.
Conclusions
MTHFR C677T had no effect on venous thromboembolism; factor V Leiden and prothrombin 20210A were confirmed as moderate risk factors.
Limitations
Observational studies published through January 2010; heterogeneity between datasets.
