Methodology
Double-blind, randomised, placebo-controlled Phase 0/1b trial (RENEWAL-NAD+) in healthy adults aged 45-75 (60 randomised, primary analysis n = 50) of 5 days of an oral NAD+ formulation (LNAD+) modified in its solution behaviour. Primary endpoints were NAD inside blood cells (whole blood) and NAD in plasma.
Key Findings
Intracellular NAD rose 53% versus placebo at day 6 (p = 5.48e-14), while plasma NAD was unchanged (p = 0.60). Plasma NAD breakdown products (MeNAM, 2PY) rose sharply. Symptom incidence was similar between groups (p = 0.68); one mild adverse event (nausea) occurred on LNAD+. No secondary clinical, vital-sign, wellbeing or wearable endpoint survived correction for multiple testing.
Conclusions
Oral LNAD+ rapidly raised NAD inside blood cells and was well tolerated in the short term.
Limitations
Five days only; one proprietary formulation, so results may not apply to standard NAD+ capsules; retrospectively registered; no clinical benefit shown; the abstract does not state the dose.
Supplements Studied
LNAD+ produced a rapid and pronounced increase in icNAD
Outcomes Measured
53% elevation versus placebo at Day 6
