Methodology
Double-blind phase I randomised trial (NADPARK) in 30 newly diagnosed, untreated people with Parkinson's disease taking 1,000 mg nicotinamide riboside or placebo for 30 days, with brain NAD measured by 31P magnetic resonance spectroscopy.
Key Findings
NR was well tolerated and produced a significant but variable rise in brain NAD and related cerebrospinal-fluid metabolites. Those whose brain NAD rose showed altered cerebral metabolism on FDG-PET, associated with mild clinical improvement. NR lowered inflammatory cytokines in serum and cerebrospinal fluid.
Conclusions
NR is a candidate neuroprotective therapy for Parkinson's disease that warrants larger trials.
Limitations
Phase I safety trial with 30 people and 30 days; clinical improvement was mild and seen in responders only; not powered for clinical efficacy.
Supplements Studied
NR treatment was well tolerated and led to a significant, but variable, increase in cerebral NAD levels
