Methodology
Randomized, double-blind, multicenter trial of acetyl-L-carnitine 1000 mg three times daily versus placebo for 24 weeks during adjuvant taxane chemotherapy, followed to 104 weeks.
Key Findings
Acetyl-L-carnitine produced statistically significantly worse neuropathy scores than placebo (-1.39 points, 95% CI -2.48 to -0.30), persisting over two years.
Conclusions
Acetyl-L-carnitine did not prevent chemotherapy-induced peripheral neuropathy and appeared to worsen it.

Supplements Studied
Patients receiving ALC had a statistically significantly (P = .01) greater reduction in NTX scores (worse CIPN) of -1.39 points (95% confidence interval [CI] = -2.48 to -0.30) than the placebo group.
