Methodology
Comprehensive systematic review and random-effects meta-analysis of randomised placebo-controlled trials of omega-3 PUFA in depressive disorders, with subgroup analyses by EPA/DHA ratio, dose and diagnostic status.
Key Findings
Meta-analysis of 11 RCTs in diagnosed MDD and 8 in depressive symptoms without MDD found significant overall benefit of omega-3 PUFA versus placebo, with effects driven by EPA-predominant formulations (>=60% EPA) at doses of 200-2200 mg/day of EPA in excess of DHA; DHA-predominant preparations showed no significant effect.
Conclusions
The use of omega-3 PUFA is effective in patients with diagnosis of MDD and on depressive patients without diagnosis of MDD.
Limitations
Substantial heterogeneity across trials; many small samples; publication bias detected on funnel plot; variable adjunctive antidepressant use; short treatment durations (mostly 4-16 weeks).
Supplements Studied
The use of omega-3 PUFA is effective in patients with diagnosis of MDD and on depressive patients without diagnosis of MDD.
