Acute Alpha-Glycerylphosphorylcholine Supplementation Enhances Cognitive Performance in Healthy Men

Methodology
Randomized, double-blind, placebo-controlled crossover trial in 20 resistance-trained men comparing placebo, 315 mg and 630 mg acute alpha-GPC, with Stroop/N-Back/Flanker testing, vertical jump and bench press throws, a Smith squat protocol and post-exercise growth hormone sampling (NCT06690619).
Key Findings
Stroop total score improved versus placebo at 630 mg (p = 0.013) and 315 mg (p = 0.046), with faster Stroop completion at 630 mg. No significant differences were found for Flanker, N-Back, visual analog scales, physical performance or growth hormone.
Conclusions
Acute alpha-GPC improved one measure of executive cognitive performance in healthy young men but did not improve physical performance or growth hormone response.
Limitations
Small single-site sample of trained men, acute single-dose design, benefit limited to one of three cognitive tests, and the author is a paid advisor to the supplement sponsor.

Outcomes Measured
When compared to PL, changes in Stroop total score were statistically greater after HD (13.0 ± 8.2 vs. 5.2 ± 9.0, p = 0.013, d = 0.61) and LD (10.8 ± 7.7 vs. 5.2 ± 9.0, p = 0.046, d = 0.48) administration.
No significant differences between groups were found for the Flanker and N-Back assessments, while a tendency was observed for HD to have faster reaction times when compared to PL during the Flanker test.
No group differences were realized for visual analog scales, physical performance, or growth hormone.
