Activity of Choline Alphoscerate on Adult-Onset Cognitive Dysfunctions: A Systematic Review and Meta-Analysis

Methodology
Systematic review and meta-analysis of PubMed, Web of Science and Embase; seven randomized controlled trials and one prospective cohort study of choline alphoscerate alone or with donepezil in adult-onset cognitive impairment, appraised with the Cochrane risk of bias tool.
Key Findings
Alpha-GPC combined with donepezil improved cognition (4 RCTs, MD 1.72, 95% CI 0.20 to 3.25), function (3 RCTs, MD 0.79, 95% CI 0.34 to 1.23) and behavior (4 RCTs, MD -7.61, 95% CI -10.31 to -4.91); patients receiving alpha-GPC had better cognition than placebo or other medications (MD 3.50, 95% CI 0.36 to 6.63).
Conclusions
Alpha-GPC alone or with donepezil improved cognition, behavior and function in patients with neurological conditions associated with cerebrovascular injury.
Limitations
Small number of included trials, mostly in clinical populations with cerebrovascular injury rather than healthy adults; several trials share the same research group; heterogeneity across measures.
