    Magnesium status and supplementation influence vitamin D status and metabolism: results from a randomized trial

    Methodology

    Randomized, double-blind, placebo-controlled trial of personalised-dose magnesium supplementation, with vitamin D metabolites measured by mass spectrometry and analysed by baseline 25(OH)D concentration.

    Key Findings

    Magnesium supplementation modified serum 25-hydroxyvitamin D in a baseline-dependent way: it raised 25(OH)D in participants whose starting concentrations were low and lowered it in those with high starting concentrations, and it also altered 24,25- and 1,25-dihydroxyvitamin D metabolites.

    Conclusions

    Our findings suggest that optimal magnesium status may be important for optimizing 25(OH)D status.

    Limitations

    Secondary analysis of a colorectal adenoma prevention trial, modest sample (n=180), and interaction findings that require confirmation in trials designed with vitamin D status as the primary endpoint.
