Methodology
Double-blind, placebo-controlled randomised trial of 1000 mcg/day oral cyanocobalamin for 12 months in older adults with moderate vitamin B12 deficiency.
Key Findings
Supplementation markedly improved biochemical B12 status but produced no significant difference versus placebo in peripheral nerve conduction, central motor conduction or cognitive test scores.
Conclusions
Twelve months of oral B12 in moderately deficient older adults corrected biochemistry without measurable neurological or cognitive benefit.
Limitations
Participants had moderate rather than severe deficiency and were largely asymptomatic; 12 months may be too short for neurological change.
